Enzymatic hydrolysis of cellulose requires mixtures of 19 hydrolytic enzymes including endoglucanases, exoglucanases (cellobiohydrolases) and 20 β-glucosidases acting in a synergistic manner. In biopolymer hydrolysis studies, 21 enzyme assay is an indispensable part. The most commonly used assays for the 22 individual enzymes as well as total cellulase activity measurements, including their 23

Strong cellulase inhibition by Mannan polysaccharides in cellulose conversion to sugars.

Cellulase enzymes contribute a major fraction of the total cost for biological conversion of lignocellulosic biomass to fuels and chemicals. Although a several fold reduction in cellulase production costs and enhancement of cellulase activity and stability have been reported in recent years, sugar yields are still lower at low enzyme doses than desired commercially. We recently reported that he...

Process relevant screening of cellulolytic organisms for consolidated bioprocessing

BACKGROUND Although the biocatalytic conversion of cellulosic biomass could replace fossil oil for the production of various compounds, it is often not economically viable due to the high costs of cellulolytic enzymes. One possibility to reduce costs is consolidated bioprocessing (CBP), integrating cellulase production, hydrolysis of cellulose, and the fermentation of the released sugars to the...

Engineering Aspergillus oryzae A-4 through the Chromosomal Insertion of Foreign Cellulase Expression Cassette to Improve Conversion of Cellulosic Biomass into Lipids

A genetic modification scheme was designed for Aspergillus oryzae A-4, a natural cellulosic lipids producer, to enhance its lipid production from biomass by putting the spotlight on improving cellulase secretion. Four cellulase genes were separately expressed in A-4 under the control of hlyA promoter, with the help of the successful development of a chromosomal genetic manipulation system. Comp...

High temperature pre-digestion of corn stover biomass for improved product yields

INTRODUCTION The efficient conversion of lignocellulosic feedstocks remains a key step in the commercialization of biofuels. One of the barriers to cost-effective conversion of lignocellulosic biomass to sugars remains the enzymatic saccharification process step. Here, we describe a novel hybrid processing approach comprising enzymatic pre-digestion with newly characterized hyperthermophilic en...
